The main concern would not be filling the hole or even the presence of the ants but the possibility that the tree may not be structurally sound. You really should have the tree looked at by a person who is experienced in assessing trees and potential hazard. Now that you can see inside the tree that may make the situation more obvious. As for getting rid of the ants, they are not the cause of the problem but a symptom of the condition of the wood in the tree, removing them would not improve the tree as it presently stands. Filling the cavity with any type of product is merely an aesthetic step, although it can be useful in closing off a cavity to further external impact, but it will not strengthen the tree if the structural strength of the tree is impaired. At this point your first concern should be the potential for failure and the potential for damage should the tree fail. Once this has been assessed, you then can decide on your course of action. If the size, location and severit
y of the cavity are sign
ificant and the tree could cause injury or damage should it fail, the tree may be best removed. If, upon assessment, the situation is considered not to present a serious concern and/or no target exists - then you can consider your other options. Have the tree looked at by a professional before you do anything else.
